Cleavage Conditions:

  • Mild cleavage: 1–2% TFA in DCM → releases protected peptide fragments

  • Strong cleavage: Standard TFA cocktail (TFA/TIS/H₂O) → fully deprotected peptide with C-terminal carboxylic acid

Applications:

  • Synthesis of fragment peptides for segment condensation

  • Generation of protected peptide intermediates

  • Ideal for peptide libraries and combinatorial chemistry
